Senescent Cell Clearance

Senescent cells, also known as “zombie cells”, are cells that have stopped dividing but remain alive. They accumulate in the body and can release harmful substances that can contribute to aging, age-related health problems, and have detrimental effects on muscle, strength, endurance, and performance.

Muscle tissue is made up of a network of cells. These cells are constantly dividing and regenerating in order to maintain the health and function of the muscle tissue. However, at a certain point, these cells may stop regenerating and become senescent – which means that the cell is no longer a functioning part of the muscle tissue, but instead of ‘dying’ and being replaced by a healthy cell, it stays there taking up space where a healthy and contributing cell could be.

The higher the number of senescent cells in a tissue, the lower rate the tissue can perform.

To help you better understand senescence and the importance of senescent cell clearance in simple terms, think of it like defragging a computer or rebooting a phone – getting rid of the clutter and things that are slowing it down and making it not perform the way it is capable of:

  • Analogy 1: Senescent cells are like old, fragmented files on your computer that have been left behind during previous tasks, causing your system to slow down and become less efficient. These files don’t contribute anything useful, but they’re still taking up space, slowing down overall performance. You defrag your computer to help speed up and optimize performance.
  • Analogy 2: You could think of senescent cells as old apps running on your phone that drain the battery and take up space, slowing down performance. Improving senescent cell clearance would be like rebooting the phone, closing the apps, and letting the system reload fresh, new software to make it work better and more efficiently.

Research has shown that BMPS can play an important role in body-composition, both in building muscle and in reducing body-fat.

Bone Morphogenetic Proteins (BMPs) – A Better Understanding

When it comes to building muscle, losing fat, improving recovery, and recovering from injuries, there is so much going on inside our bodies that we can’t see with the naked eye.

A group of powerful proteins known as Bone Morphogenetic Proteins (BMPs) plays a crucial role in many of these processes. While BMPs were originally recognized for their role in bone formation and healing, newer research has shown that they can also promote muscle growth, improve fat metabolism, and speed up recovery.

What are Bone Morphogenetic Proteins (BMPs)?

Bone Morphogenetic Proteins (BMPs) are a group of growth factors that belong to a family of proteins called TGF-beta (transforming growth factor-beta).

While the name “Bone Morphogenetic” suggests their primary function is related to bones, these proteins are involved in more than just bone growth—they play significant roles in muscle tissue repair, fat metabolism, and healing after injury.

Bone Morphogenetic Proteins (BMPs) have been found to play a crucial role in muscle anabolism, promoting muscle growth and inhibiting muscle wasting.

In simple terms, BMPs act as ‘messengers’ that tell cells in our body how to behave. They help:

  • Build and maintain bone tissue.
  • Stimulate the formation of muscle cells and promote muscle repair.
  • Influence the way the body stores and burns fat.
  • Aid in tissue recovery after injury, both for bones and muscles.

Important definitions:

  • BMP Signaling – refers to the process of how BMPs activate cellular responses through their receptors and intracellular signaling pathways.
  • BMP Levels – refer to the actual concentration of BMPs, which can modulate the intensity or outcomes of BMP signaling.
  • BMP Expression – refers to the genes activated by the signaling pathways.

While BMPs are involved in many processes in the body, their impact on muscle building, fat loss, and recovery is especially important for anyone looking to improve their fitness, health, or athletic performance.

Key BMP’s Involved in Muscle Growth, Fat Loss, & Recovery

There are over 20 different bone morphogenetic proteins, each with different roles in the body.

Some of the most important BMPs when it comes to building muscle, losing fat, and recovery are:

  • BMP-2: BMP-2 is considered a potent growth factor that can significantly support enhanced bone regeneration by stimulating the differentiation of stem cells into osteoblasts (bone-forming cells) and promote new bone formation. In addition, research suggests that it can also play a key role in muscle growth and regeneration, potentially promoting muscle growth and inhibiting muscle wasting.
  • BMP-4: While BMP-4 is primarily known for its role in bone development, it also plays a role in muscle building, particularly in regulating the proliferation and differentiation of satellite cells, which are the stem cells of adult skeletal muscle. It is also important for muscle repair and regeneration and some studies have shown that BMP-4 signaling may downregulate myostatin expression.
  • BMP-7: BMP-7 plays an important role in muscle growth, with studies showing its ability to promote hypertrophy (muscle building) and to protect against atrophy (muscle wasting). It is known for its role in supporting healing in bones and tissues, and is involved in muscle regeneration, particularly after muscle injuries. It has been shown to stimulate the differentiation of myoblasts into mature cells, which is important for repairing damaged muscle fibers and helping to restore healthy muscle function. It may also help prevent fibrosis (scar tissue formation) during muscle tissue repair, which is significant because fibrosis can impede muscle regeneration and lead to loss of functionality. Studies have also shown that increasing BMP-7 signaling can increase fat oxidation and energy expenditure (burning calories).

With that in mind, let’s look at which ingredients in BASIS have been shown to increase the levels, signaling, or expression of:

It’s important to emphasize that even if you have taken some of these ingredients individually by themselves in the past, the real power is taking them together in order to effectively target the different BMP pathways together.

Let’s Examine the Ingredients

In the section above, we listed out which ingredients are thought to work via which Bone Morphogenetic Protein (BMP) pathways, but there’s great news – because as great as it is that BASIS works through key BMP pathways, there’s a lot more to BASIS!

Almost every ingredient in BASIS is included because it helps support building muscle &/or burning fat via more than one pathway, and in most cases, these pathways come together to compliment each other and help provide increased synergistic results!

Icariin (from Horny Goat Weed) (as HG20X™) – 500 mg. HGW/100 mg. Icariin

Icariin is derived from Horny Goat Weed (Epimedium). There are numerous standardizations available, but for the purposes of inclusion in BASIS, we selected the ultra-potent HG20X™ 20% Icariin extract.

This is a great example of an ingredient that has become so well-known for one subject or category that people often overlook any other potential benefits that it may have.

The Importance of Standardization (Icariin)

As with many herbs, it’s not the herb itself that provides the benefits, but the active constituent that the herb is standardized for.

The active constituent of Horny Goat Weed (Epimedium) that provides the benefits is Icariin.

The active ingredient related to the bone morphogenetic protein aspect is Icariin.

So what matters is the amount of Icariin in the product.

That’s why we use HG20X™, a Horny Goat Weed standardized for a minimum 20% Icariin.

Beware – Icariins and Icariin are Very Different

It is very important to distinguish between Icariin and Icariins, as the (s) at the end makes all the difference. You want Icariin without the (s) at the end, as it is Icariin that provides the benefits whereas Icariins do not.

Unfortunately, many Horny Goat Weed products on the market are not standardized at all or are standardized for Icariins, as that is a lot cheaper than being standardized for Icariin. This allows for brands to significantly cut product costs, but is deceptive towards consumers, as most consumers don’t know the difference and what they should be looking for.

Sexual Health & Libido

Icariin supports an increase in natural testosterone levels and libido/sex drive and has also been shown to support improved erection quality by acting as a natural PDE5 inhibitor, meaning that it may help inhibit the protein phosphodiesterase 5 enzyme that interferes with blood flow to the penis.

Enhanced Nitric Oxide/eNOS Production

Icariin has been shown to increase nitric oxide (NO) production by stimulating endothelial nitric oxide synthase (eNOS) and activating signaling pathways like PI3K/Akt. This plays a role in promoting blood flow and nutrient delivery to muscle tissues and can support improved muscle endurance, faster muscle recovery, and increased muscle fullness, pumps, and vascularity.

Icariin and Bone Morphogenetic Protein

Icariin is the main active flavonoid glucoside from the Chinese Epimedium herb, better known in some parts of the world as Horny Goat Weed. Epimedium has a long history of use in traditional Chinese medicine (TCM) for osteoporosis, osteoarthritis, and bone fractures.

Icariin has been shown in studies to have positive benefits on bone metabolism, including anti-osteoporosis, anti-osteoclastogenesis, and osteogenesis (the formation of bone).

Several studies have shown that Icariin can increase osteogenic differentiation and reduce bone loss in vivo and in vitro. Osteogenic differentiation is a crucial biological process for maintaining bone remodeling and fracture repair.

Important Reminder: Although Bone Morphogenic Protein (BMP) sounds like it would be specifically related to bones, researchers have found that BMP can be a limiting factor as to how much muscle growth one can achieve, making stimulating BMP an exciting new pathway to increasing muscle and strength and improving body-composition.

Studies have shown that icariin treatment increases the expression of BMP-2, BMP-4, and BMP-7.

The pathways through which Icariin has been shown to support BMPs include a variety of critical signaling pathways such as PPARγ, ERα/AKT/β-catenin, and MAPK, and many researchers believe that there are still a lot of underlying mechanisms that have yet to be fully understood.

Icariin has also been shown to promote BMP2 by activating the cAMP/PKA/CREB signaling axis pathway. Researchers then used a PKA selective inhibitor to inhibit the effect of Icariin on osteogenesis, thus reinforcing that Icariin was indeed having a stimulatory effect on BMP2 and osteogenesis.

Here is an excerpt from one study that confirms Icariin increases BMP-7, inhibits PDE5, and has promising potential for fat loss due to these mechanisms:

  • Study: Icariin has the potential to induce the differentiation of bone marrow mesenchymal stem cells into brown fat cells via PDE5A inhibition
  • Excerpt: Bone morphogenetic protein-7 (BMP-7) is reported to induce the PGC-1α and elevate the expression of UCP-1 and PPAR-γ to activate brown adipogenesis. In addition, the WAT browning is accompanied by UCP-1 upregulation, increasing the energy consumption, and attenuating the inflammatory response in adipose tissues.
  • Conclusion: The results of this study demonstrated that ICA could induce the differentiation of BMMSCs into brown fat cells via PDE5A inhibition, suggesting that ICA may become the promising therapeutic agent for obesity related diseases.

In simple terms, this study demonstrates that:

  • Icariin does increase BMP-7.
  • Icariin does inhibit PDE5.
  • Icariin is shown to have promising potential for fat loss.

In summary, Icariin is included in BASIS to help support an increase in BMP-2, BMP-4, and BMP-7 expression and levels, its potential role in increasing nitric oxide & eNOS, and for its natural PDE5 inhibition support benefits.

Hwanggeumchal Sorghum Extract – 500 mg.

Hwanggeumchal Sorghum Extract has been shown to enhance BMP-7 and Growth Hormone (GH) signaling through the activation of the Jak2/STAT5B pathway, a crucial pathway in GH signaling.

Growth Hormone (GH) has a well-established influential effect on bone growth, muscle mass, and body-composition. When GH levels are elevated, it triggers something called the Jak2/STAT pathway which regulates IGF-1, which is another major player in bone and muscle growth.

Hwanggeumchal sorghum extract has been shown to act almost exactly like Growth Hormone in activating the Jak2/STAT pathway, which then increases the expression of GH related proteins, the GH receptor, IGF-1, the IGF-1 receptor, and BMP7. This can help influence an optimal muscle building environment.

Hwanggeumchal Sorghum extract has also been the subject of some very interesting anti-aging and longevity research.

Naringin – 250 mg.

Naringin is a natural citrus bioflavonoid found in citrus fruits that has been researched for a wide variety of potential benefits. It has been shown in studies that it may have antioxidant, anti-inflammatory, cardioprotective, and neuroprotective benefits.

Naringin has been researched to see if it would help with metabolic syndrome, and was shown to support reducing blood pressure, glucose levels, and fasting insulin levels.

Naringin has also been shown to increase absorption of and the effects of some other ingredients.

Bone Morphogenetic Protein Research

Osteoporosis is the reduction in skeletal mass due to an imbalance between bone resorption and bone formation. Bone morphogenetic protein (BMP) plays an important role in osteoblastic differentiation and bone formation. Naringin has been researched for its role in BMP activation.

Bone and cartilage disorders are the leading causes of musculoskeletal disability. There is no cure for all of these disorders, so they are the subject of a great amount of research, including on natural compounds. Naringin has been researched due to its potential multifaceted activities in bone and cartilage tissue.

There has been an immense amount of information about Naringin learned from these studies.

It is important to note that the information in this write up is provided to show how Naringin may support an increase in bone morphogenetic protein levels and that no claims are being made about osteoporosis or bone or cartilage conditions. Any mention of osteoporosis &/or bone and cartilage conditions is only mentioned to provide the context of the study findings as they may relate to bone morphogenetic proteins.

Studies indicate that Naringin may promote osteogenic differentiation, inhibit osteoclast formation, and exhibit protective effects against osteoporosis and in bone and cartilage disorders in vivo and in vitro. Many signaling pathways, such as BMP-2, Wnt/β-catenin, and VEGF/VEGFR, participate in the biological actions of Naringin in mediating the pathological development of osteoporosis.

One study conclusion noted:

Further biological and pharmacological evaluations of naringin and its applications in bone tissue engineering, particularly its therapeutic effects against osteoporosis, might result in developing potential drug candidates.

Another study noted:

Our results provide evidence that Naringin increases BMP-2 expression and enhances osteogenic response via the phosphoinositide 3-kinase (PI3K), Akt, c-Fos/c-Jun and AP-1-dependent signaling pathway.

To Paraphrase on the Matter of Naringin and Bone Morphogenetic Protein: 

Naringin has been shown in studies to support increases in mRNA and BMP-2 levels and to help reduce the decreasing of BMP-2 and bone loss.

Bone Morphogenetic Protein

Kaempferol has been shown to increase BMP-2 and BMP-7 expression, likely by activating the ERK/BMP-2 signaling pathways, which increases the expression of BMP-2 and BMP-7. These BMPs are involved in tissue regeneration and repair, including muscle tissue. By promoting the differentiation and regeneration of muscle cells, Kaempferol could aid in muscle recovery and muscle growth.

Kaempferol has been found to activate several intracellular signaling pathways that regulate gene expression. For example, research suggests that Kaempferol can upregulate MAPK signaling pathways, which can regulate BMP signaling, and which play a crucial role in regulating various cellular processes, including muscle growth and metabolism.

Kaempferol induced autophagy (a cellular process that removes damaged components) may also contribute to osteoblast differentiation and help maintain healthy muscle cells, further supporting muscle building efforts. (This ties in with the Apigenin and Senactiv® as well).

Ligusticum Wallichii Extract (98% Ligustrazine) – Soy Free – 200 Mg.

Ligustrazine is a natural compound found in the roots of the Asian plant Ligusticum Wallichii.

Ligustrazine has potential anti-inflammatory and nootropic benefits, but for the purpose of this write up we are focused on its potential muscle building and muscle sparing benefits.

Ligustrazine has been shown to support the elevation of BMP-7, a potent anti-inflammatory growth factor. It is also thought to support a reduction in the activity of the catabolic signal molecule MuRF1 and has been shown in multiple studies to reduce the amount of muscle loss as compared to control groups, in both short-term and longer-term studies.

Ligustrazine has also been shown to support an increase in glucose uptake to muscle cells and to help protect against the oxidative damage from both high fat and high sugar diets. It has also been shown to support an increase in fatty acid utilization. This not only may increase fat burning but may also spare glucose to be used in the muscle cells to preserve energy & muscle mass.

Ligustrazine has even been shown to support improved mitochondrial biogenesis.

Osthole Cyclodextrin Complex – 200 mg.

Osthole is a dietary supplement ingredient that has been used to support cardiovascular health, cognitive health, liver health, vasodilation, optimal hormone levels, and has natural PDE5 inhibition properties.

Natural PDE5 Inhibition Properties

Cnidium Monnieri, the herb that Osthole is derived from, and Osthole itself, have both been shown in research to have PDE5 inhibitor properties. Apigenin is an exciting natural muscle building, general health, healthy aging, & longevity phytonutrient.

Apigenin is a natural flavonoid found in parsley, celery, chamomile, and citrus fruits and is similar to Quercetin, Hesperidin, Luteolin, and Kaempferol. However, even though its structure is very similar to these compounds, Apigenin has some very different effects in the body than those related substances.

Apigenin has a natural anabolic effect, as first discovered by researchers at the Korea Food Research Institute. Researchers found that it has the unique ability to inhibit 17β-hydroxysteroid dehydrogenase (17β-HSD) and animal studies have shown it to support muscle protein synthesis and muscle growth via increased phosphorylation of several key players in the mTOR pathway including Akt, S6K1, & 4E-BP1.

In the initial study that led to Apigenin’s popularity for building muscle, scientists began experimenting with this flavone with mice and the mice receiving the Apigenin had significant muscle mass gains over the mice not receiving it. The greater the dosage of Apigenin the mice took, the larger the quad increase!

Even before the research showing its natural anabolic benefits, Apigenin was already regarded as a very exciting compound due to its wide variety of overall health benefits including its anti-inflammatory benefits as well as its potential prostate, neuroprotective, and cardioprotective properties.

Another very exciting aspect of Apigenin is its ability to enhance autophagy. Autophagy is a natural cellular mechanism by which by which the cells in our body degrade unnecessary or damaged components within the cell. The process of autophagy helps maintain normal functioning (homeostasis) in the cell (think back to the defragging a computer analogy in the senescence section).

Apigenin & Bone Morphogenetic Proteins (BMPs)

Apigenin has been shown to influence several biological pathways, including Bone Morphogenetic Protein (BMP) levels, signaling, and expression.

Activation of BMP Pathways

Apigenin has been shown to support the activation of BMP signaling pathways, likely through the upregulation of BMP receptors and ligands. Apigenin can influence the modulation of the expression of genes involved in BMP signaling, possibly by affecting transcription factors or intracellular signaling molecules that regulate these processes.

Modulation of Gene Expression

Apigenin may influence the expression of BMP-related genes. For instance, studies have shown that Apigenin may upregulate the expression of BMP-2 and BMP-4. This upregulation can occur via the activation of transcription factors, such as SMAD proteins, which are key mediators in the BMP signaling cascade.

BMP Receptor Activity

BMP receptors play a central role in the activation of the BMP signaling pathway. Apigenin may enhance the sensitivity of cells to BMP signaling by increasing the expression or activity of these receptors, facilitating the downstream effects of BMP signaling, which could lead to a stronger activation of the pathway.

SMAD Pathway Activation

The SMAD signaling pathway is one of the downstream effects of BMP receptor activation. Apigenin can promote the activation of SMAD1/5/8 phosphorylation, which then translocates to the nucleus to regulate the transcription of target genes involved in bone formation. This suggests that apigenin may enhance BMP-mediated osteogenesis by activating the SMAD pathway.

Inhibition of Negative Regulators

Apigenin may also support an increase in BMP signaling by inhibiting negative regulators such as Noggin and Chordin, which both act to suppress BMP activity. By supporting a reduction in the expression of these inhibitors, Apigenin may promote a more robust BMP signaling response.

Osteogenesis Enhancement Support

Apigenin has been noted to support the enhancement of osteogenic differentiation in mesenchymal stem cells (MSCs). This is likely due to the combined effects of increasing BMP signaling and promoting the expression of osteogenic markers, which are crucial for bone formation and regeneration.
